You should specify that we're talking about main characters, Gravity has a few characters in the beginning, and My Dinner With Andre has the waiter.

if we are talking 'two main characters', then we have to include my favorite film of all time...'chinese coffee' starring jerry orbach and al pacino...

a brilliant movie that started out as a two man play...(the movie just throws in a few quick unnecessary flashbacks)...

Ooh, I LOVE minimalist movies like that, so behold my pile of suggestions ranging from no budget efforts to real masterpieces of craft. I'd love to a few movies like that myself, but making them right actually require professionalism and experience. Ironically enough, the less is going on the harder is to make the movie.

The Caller (1987) (two characters; virtually unknown cult masterpiece, a great suspenseful thriller with one of the best endings ever)

Tayna Slendermena (2013) (two characters)
Kollektor (2016) (one character, very similar to Locke)
The Last Days of Earth (2017) (okay, I am actually not sure whether this San-Marino/Ukraine no budget co-production starring yours truly counts ... All of the discernible characters were portrayed by me, but I'm not entirely sure whether I was portraying three different people or all of them are one. I guess that is left to interpretation)
Cursed Forest (2018) (one and a half character)
Chaos (2019) (two characters; one of them is portrayed by me)

as did Robert Prosky who also was nominated for the 1988 Tony Award for Best Actor in Play.
