The Duke is one of my all time favorites.  I don't know how many you want from each of us, but I'll give you three (in order).

One of his toughest characters in a western is J.B. Books in The Shootist.  It was Wayne's last movies.  The character knows that he is dying of cancer, but still finds time to help others.

My second favorite tough guy in a western would be U.S. Marshall Rooster Cogburn in the REAL version of True Grit.  "Fill your hands you son of a b---h!"  Mean, old, drunk, cantankerous... and fun!

Third would be 1963's McClintock.  "I haven't lost my temper in forty years, but pilgrim you caused a lot of trouble this morning, might have got somebody killed... and somebody oughta belt you in the mouth. But I won't, I won't. The hell I won't!"  The scene where he tracks down Maureen O'Hara and then spanks her is one of the funniest ever!
