Mind me shaking my fist at you for making a solid poll suggestion that's pretty impossible to nominate options due to ... nonexisting keywords. Not a critique against you but my own poor memory recall. I'm not sure what movies I've seen (or know about) that would qualify. I must have over the many decades watched something good that would fit here. Damned Google isn't a help.
Honorable mention for Metropolitan (1990) which I think has five main characters. "The cynic Nick, the philosophical Charlie, party girl Sally and austenite Audrey. They are joined by Tom.
Dishonorable mention: The Bobbsey Twins.
The Bobbsey Twins are made up of the Bobbsey siblings, which are two sets of fraternal twins: 12-year-old Nan and Bert, and 6-year-old Flossie and Freddie.

It's great to see three Mike Nichols films on the list. I feel like I have some suggestions but they're not coming to me. My vote will go to 'Closer,' which I think is under-rated, followed by 'Sex, Lies, and Videotape' and 'Virginia Woolf.'

Sorry for being so late to reply, I've added pretty much all the suggestions, except for the horror or sci-fi movies. Here's why; this poll focuses on movies whose main purpose is to comment on human relationships based on friendship, love, sex etc. and then using two couples for that. That's why I added "relationship-themed", so we're pretty much dealing with dramas or comedies (or comedy-dramas).
If you feel that one of your suggestions don't match anymore, tell me so and I'll remove it.
And yes, Ruby, Mike Nichols seems to be the master of the genre :)
